课题名称:斑节对虾海产品绿色安全养殖生产的生物探讨

(一)课题技术点及分析:

斑节对虾(Penaeus monodon):1)为可人工养殖的对虾的一种。2)弧菌数量:当其增到一定数量时,即有引发斑节对虾发病的危险。3)蛭弧菌(Bdellovibrio):为一类寄生于细菌体内的更小的细菌。它可以通过裂解弧菌而达到控制弧菌数量的目的。

本课题将采用生物的手段,探讨在斑节对虾绿色安全养殖生产过程中应用的可能。也就是说,在不加任何化学药物、抗生素等可能导致药残超标的情况下,采用蛭弧菌有益菌,以控制斑节对虾养殖过程中的有害弧菌的数量,进而控制其病害的发生,最终获得无药残的绿色海产品。

技术特征为斑节对虾养殖过程中的弧菌数量的生物(蛭弧菌微生态制剂)控制。

[文摘] 研究了施用10 mg·kg^-1光合细菌以及1、2和3 mg·kg^-1芽孢杆菌制剂对斑节对虾亲虾培育池水体异养细菌和弧菌数量的影响.结果表明:在试验初期,试验组和对照组水体总异养细菌和弧菌数量都明显上升;试验后期,试验池水体异养细菌和弧菌数量均明显下降,明显低于对照池;施用10 mg·kg^-1光合细菌能显著抑制亲虾池水体异养细菌的生长(P<0.01);施用3 mg·kg^-1芽孢杆菌对亲虾池水体弧菌数量的抑制效果最好(P<0.01).

摘要:本发明是指一种高密度蛭弧菌游泳体的发酵培养技术,其技术包括如下步骤:1)将宿主菌摇床培养,离心后加入DNB液后得宿主菌悬浮液;2) 在DNB液中加入宿主菌悬浮液,再接入蛭弧菌斑后培养,离心后加入DNB 液即得蛭弧菌浓缩液;3)往DNB液中先加入谷氨酸钠,再接入宿主菌悬浮液,最后接入蛭弧菌浓缩液,使其浓度为1~103PFU/mL,然后置于发酵罐中培养4~6天,即得到密度为108~1014PFU/mL的蛭弧菌发酵液体,离心浓缩即得蛭弧菌游泳体浓缩液。本发明成本低,蛭弧菌终浓度高,应用范围广,使其不但在水源性、食源性致病菌防治方面,而且在医学、环境污染、农业、工业、军事领域等方面均可使用。

摘要:一种噬菌蛭弧菌制剂的生产方法,包括如下步骤:1)将红假单胞菌混悬液与 0.2%~0.85%NaCl溶液制成含菌量为30~300亿/ml的宿主菌混悬液;2)将pH6.5~8.0的磷酸盐缓冲液用水10倍稀释,再加入CaCl2和MgCl2使所得溶液中两者含量均为 10~100mg/L,制成培养液;3)在该培养液中加入与其体积比为5%~30%的宿主菌混悬液,再加入与已加宿主菌混悬液的培养液体积比为2%~3%噬菌蛭弧菌液,在28~30℃温度条件下培养72~120小时,即得成品。本发明解决用大肠杆菌作为宿主菌很难进行工业化生产及环境污染问题,保证了噬菌蛭弧菌的裂解活性,提高了其防治病害的效果。

噬菌蛭弧菌(Bdellovibrio bacteriovorus,以下简称蛭弧菌)是一类以黏附、裂解宿主菌方式使自身得以生长繁殖的微生物,广泛存在于各种自然水体。论文试验期间在海南对当地方斑东风螺、南美白对虾、九孔鲍鱼、斜 6

带石斑鱼、点带石斑鱼、锯缘青蟹的育苗池和养殖池,绿藻藻池以及养殖区周边的海域和远离养殖区的外海海域进行海水微生物环境调查;同时对黄浦江河水、生活污水及上海沪东污水处理厂水样进行淡水微生物环境调查。调查结果发现所有被检测水域均有噬菌蛭弧菌的存在,数量在10<'1>~10<'3> pfu/ml不等,表现为与水体中细菌总数、弧菌数成正相关。 从海南琼海对虾养殖场及其周边排水沟采集水样,比较不同分离方法、上层培养基营养盐浓度和富集培养与直接培养等对海水蛭弧菌分离、计数的影响,从而建立了海水蛭弧菌分离和纯化的方法--下层1.0%琼脂,上层0.5%琼脂的无营养盐海水双层琼脂平板法。应用无营养盐的海水双层琼脂平板法,采用6种宿主菌分别对海南4份海水样品进行计数检测,发现不同宿主菌所得到的蛭弧菌数量不同,根据噬菌斑的大小、形状和斑形成速度分离出235株蛭弧菌海水菌株,同时对分离纯化到的3株蛭弧菌海水菌株进行液体培养增殖,进一步检验了海水双层琼脂平板法的可靠性。 对利用双层琼脂平板法分离纯化获得的蛭弧菌海水菌株.Bdh5221 裂解特性及生长影响因子进行了初步研究,研究结果表明,蛭弧菌海水菌株:Bdh5221对嗜水气单胞菌、副溶血弧菌、鳗弧菌、溶藻弧菌、哈维氏弧菌、大肠杆菌、荧光假单胞菌、迟钝爱德华氏菌等革兰氏阴性菌有较好的裂解作用,对八叠球菌、金黄色葡萄球菌、枯草芽胞杆菌等革兰氏阳性菌的裂解能力较弱,对木糖葡萄球菌和啤酒酵母菌不裂解;可在超声波破碎后的G<'+>菌和酵母菌上生长。蛭弧菌Bdh5221在宿主菌浓度10<'6>~10<'9>cfu/ml下均可正常生长,高浓度宿主菌更利于蛭弧菌生长;Bdh5221最适生长的pH在7.0~7.5之间,盐度为20~30左右;Bdh5221增殖速度与起始接种浓度影响不大,起始接种浓度10<'1>~10<'3>pfu/ml下经过5~7d生长可增殖4log左右;Bdh5221保藏选择20%~30%甘油含量较为合适,低温条件更利于保藏。 安全性试验结果表明,添加1.5×10<'6>pfu/ml蛭弧菌海水菌株Bdh5221对南美白对虾无毒害作用;添加2.0×10<'2>pfu/ml,2.0×10<'3>pfu/ml,2.0×10<'4> pfu/mlBdh5221均对锯缘青蟹蚤状幼体阶段五个变态期幼体存活率和变态率无影响。锯缘青蟹苗种培育中应用蛭弧菌海水菌株Bdh5221试验,进一步表明,其最适有效添加量为1.5×10<'1>pfu/ml;添加蛭弧菌的试验组水体微生态环境明显改善,在育苗后期,相较对照组而言,试验组水体中弧菌和细菌总数都维持在较低水平,两者相差1~2个数量级,而成活率和变态率试验组则比对照组分别高6.6%和6.3%;同时试验组水质有所改善,其中NH<,4>-N比对照组低0.11~0.34 mg/1。 从蛭弧菌在锯缘青蟹苗种培育的应用试验中可以看出,其在海水苗种培育中维持微生态平衡,防治病害方面有广泛的应用前景。

摘要:水产养殖易受各种病原微生物的侵害,而各种抗生素及其他化学药物的使用势必造成二次污染。海洋蛭弧菌微生态制剂是利用该菌对水产动物致病性宿主菌天然的捕食及净化特性来进行水产养殖动物病害防治。该项目以水产动物体分离到的病原弧菌为宿主,从海水中分离出一株高活性菌株,利用该菌发酵液活菌制成纯天然的生物制剂。该制剂产品可应用于水产养殖细菌性疾病防治及促进生长。该微生态制剂的活菌浓度达到10^8Pfu/ml以上,不含杂菌,其急性安全性实验结果显示无毒,在实验室条件下对细菌的杀灭率达到98.85%,在对虾育苗试验中使对虾育苗成活率平均提高了33.6%。主要应用在水产养殖细菌性疾病的预防与治疗及促进生长。

Abstract

Four marine Bdellovibrio-and-like organisms (BALOs), viz. BDW01, BDW02, BDW03 and BDW04, were isolated from the Shenzhen Bay of Guangdong province, China, using Vibrio parahaemolyticus (Vp) strain minus as the host bacterium. They were identified as BALOs by electron microscopy and specific polymerase chain reaction methods. Lysis expts. on 41 potentially pathogenic vibrios showed that individually, BDW01, BDW02, BDW03 and BDW04 lysed 36.6, 39.0, 65.8 and 63.4% vibrios, resp. In combination, they lysed 87.8% of the vibrios tested. In terms of different vibrio species lysed, four BALOs combined could lyse 81.8% Vibrio alginolyticus (11 strains), 83.3% non-O1 Vibrio cholerae (6 strains), 88.9% Vibrio fluvialis (9 strains), 100% Vibrio mimicus (4 strains) and 88.9% Vp (9 strains). The results of this work clearly demonstrated the potential of BALOs in lysing potentially pathogenic vibrios

were found to be 10-103 PFU ml-1 in the surface water samples. A total of 130 BALOs isolates were differentiated into five phylotypes by denaturing gradient gel electrophoresis targeting the 16S rDNA V3 region and four phylotypes by amplified rDNA restriction anal. of the Bacteriovoracaceae-specific 16S rDNA fragment resp. Phylogenetic anal. of representative isolates showed that all of them were identified as Bacteriovorax spp., but affiliated with four different clusters in the family Bacteriovoracaceae. Conclusions: The two PCR-based methods both can be chosen to rapidly type the saltwater BALOs at cluster level. And the relatively large nos. of BALOs with various phylotypes recovered from the same habitats suggested that these predators might play important ecol. role in shrimp mariculture environments. Significance and Impact of the Study: We proposed two effective methods to distinguish rapidly large nos. of BALOs isolates and our results would be helpful to understand the diversity and function of BALOs in mariculture environments.

Abstract

The addn. of Timsen (alkyl di-Me benzyl ammonium chloride-40-active) to a shrimp culture pond effectively prevented the occurrence of vibriosis in shrimp. The results indicated that Timsen could effectively reduce the no. of total heterotrophic bacteria in water samples, but its efficacy was reduced in brackish as compared with fresh water.

A significant redn. of the no. of Vibrio species in brackish water samples was also obsd. In the hepatopancreas of shrimp collected from a control pond, a rapid increase in the no. of Vibrio was obsd. with the onset of disease. With prolonged exposure (120 h), Timsen still maintained lower Vibrio nos. in brackish water samples, with no difference in total nos. of heterotrophic bacteria between control samples and treated brackish water samples. Further anal. of the compn. of the Vibrio species suggested that Timsen also caused a change in the components of the Vibrio community. Results also indicated that Timsen may affect the hemocytic activity of shrimp by enhancing the activity of phagocytosis and/or encapsulation; however, it also induces hemocyte death via apoptosis.

Abstract

AIMS: The present work aims at finding potential probionts from marine sources as a biocontrol agent against pathogenic Vibrio species in shrimp larval culture. METHODS AND RESULTS: A total of 109 bacterial strains were isolated from seawater, sediment and marine fish-gut samples, and were screened for their antagonistic activity against Vibrio species. Three strains (Q, Q1 and M) isolated from the marine sediment were found antagonistic against Vibrio strains. Based on 16S ribosomal DNA gene sequence analysis, the strain Q was identified as 10

Paenibacillus spp. (EF012164); Q1 as Bacillus cereus (DQ915582); and the M as Paenibacillus polymyxa (DQ915580). Further, the two bacterial species, Paenibacillus spp. and B. cereus were challenged separately at two different concentrations of 10(4) and 10(5) CFU ml(-1) for probiotic activity in the postlarvae of Penaeus monodon against pathogenic Vibrio harveyi and Vibrio spp. CONCLUSIONS: The present study identified the probiotic activity of Paenibacillus spp., B. cereus and Pa. polymyxa against the pathogenic Vibrios in the postlarvae of P. monodon. SIGNIFICANCE AND IMPACT OF THE STUDY: In vivo study reveals that the marine bacterial species can be used as probionts against pathogenic Vibrios in shrimp larval culture practices.

Abstract

Use of antibiotics for the control of bacterial diseases in shrimp culture has caused several adverse impacts to the industry. This has resulted in the search for alternative environment friendly approaches to overcome bacterial infections. This study was conducted to investigate the use of beneficial bacteria as an alternative to antibiotics. Ten pathogenic bacterial species isolated from shrimp, Penaeus monodon, and Artemia cysts were tested for susceptibility to indigenous marine Bacillus subtilis AB65, Bacillus pumilus AB58, Bacillus licheniformis AB69 and compared with oxytetracycline, chloramphenicol, gentamicin and bacitracin, which are common antibiotics used in Asian aquaculture. The Bacillus spp. were isolated from the local marine environment for bioremediation use in shrimp hatcheries and were proven to reduce total ammonium nitrogen. The pathogenic bacterial isolates were 90% susceptible to B. subtilis AB65, 70% susceptible to B. pumilus AB58 and B. licheniformis AB69 and 100% susceptible to oxytetracycline, chloramphenicol and gentamicin but only 40% to bacitracin. Two representative isolates of the vibrio group, Vibrio alginolyticus VaM11 and Vibrio parahaemolyticus VpM1, when tested for competitive exclusion by a common broth method using the marine Bacillus spp., showed decreased viable counts from 108 to 102 cfu mL-1. The results suggest that the action of the marine bacteria appears to be significant in protecting the host shrimp against pathogenic bacteria. In addn. to the alternative use of antibiotics, the selected marine bacteria had addnl. bioremediation properties of reducing ammonia.

USE/ADVANTAGE-The Vibrio infectious disease can be treated without generating resistant microbe nor damaging safety by using Bdellovibrio (i.e. Leptospira interrogans of Vibrio). Handling of the therapeutic agent is very easy and the cost is economical compared to conventional agent.

Intramuscular injection of Vibrio, Bdellovibrio and their mixt. (0.5 ml of a sterilised sea water suspension in Vibrio and Bdellovibrio (10 cells/ml)) was carried out on Peneus japonicus (11-13g) abdomen, and also only the sterilised sea water was injected as a control. Mixing rate of the Vibrio and Bdellovibrio was 9:1. The next day, all of the 12

prawns (5) in Vibrio-injection area were dead, however, in the Vibrio-Bdellovibrio injection area, three were alive. In the control and Bdellovibrio injection areas, all 5 were alive.

摘要:水产养殖易受各种病原微生物的侵害,而各种抗生素及其他化学药物的使用势必造成二次污染。海洋蛭弧菌微生态制剂是利用该菌对水产动物致病性宿主菌天然的捕食及净化特性来进行水产养殖动物病害防治。该项目以水产动物体分离到的病原弧菌为宿主,从海水中分离出一株高活性菌株,利用该菌发酵液活菌制成纯天然的生物制剂。该制剂产品可应用于水产养殖细菌性疾病防治及促进生长。该微生态制剂的活菌浓度达到10^8Pfu/ml以上,不含杂菌,其急性安全性实验结果显示无毒,在实验室条件下对细菌的杀灭率达到98.85%,在对虾育苗试验中使对虾育苗成活率平均提高了33.6%。主要应用在水产养殖细菌性疾病的预防与治疗及促进生长。

摘要:噬菌蛭弧菌(Bdellovibrio bacteriovorus,以下简称蛭弧菌)是一类以黏附、裂解宿主菌方式使自身得以生长繁殖的微生物,广泛存在于各种自然水体。论文试验期间在海南对当地方斑东风螺、南美白对虾、九孔鲍鱼、斜带石斑鱼、点带石斑鱼、锯缘青蟹的育苗池和养殖池,绿藻藻池以及养殖区周边的海域和远离养殖区的外海海域进行海水微生物环境调查;同时对黄浦江河水、生活污水及上海沪东污水处理厂水样进行淡水微生物环境调查。调查结果发现所有被检测水域均有噬菌蛭弧菌的存在,数量在10<'1>~10<'3> pfu/ml不等,表现为与水体中细菌总数、弧菌数成正相关。 从海南琼海对虾养殖场及其周边排水沟采集水样,比较不同分离方法、上层培养基营养盐浓度和富集培养与直接培养等对海水蛭弧菌分离、计数的影响,从而建立了海水蛭弧菌分离和纯化的方法--下层1.0%琼脂,上层0.5%琼脂的无营养盐海水双层琼脂平板法。应用无营养盐的海水双层琼脂平板法,采用6种宿主菌分别对海南4份海水样品进行计数检测,发现不同宿主菌所得到的蛭弧菌数量不同,根据噬菌斑的大小、形状和斑形成速度分离出235株蛭弧菌海水菌株,同时对分离纯化到的3株蛭弧菌海水菌株进行液体培养增殖,进一步检验了海水双层琼脂平板法的可靠性。 对利用双层琼脂平板法分离纯化获得的蛭弧菌海水菌株.Bdh5221 裂解特性及生长影响因子进行了初步研究,研究结果表明,蛭弧菌海水菌株:Bdh5221对嗜水气单胞菌、副溶血弧菌、鳗弧菌、溶藻弧菌、哈维氏弧菌、大肠杆菌、荧光假单胞菌、迟钝爱德华氏菌等革兰氏阴性菌有较好的裂解作用,对八叠球菌、金黄色葡萄球菌、枯草芽胞杆菌等革兰氏阳性菌的裂解能力较弱,对木糖葡萄球菌和啤酒酵母菌不裂解;可在超声波破碎后的G<'+>菌和酵母菌上生长。蛭弧菌Bdh5221在宿主菌浓度10<'6>~10<'9>cfu/ml下均可正常生长,高浓度宿主菌更利于蛭弧菌生长;Bdh5221最适生长的pH在7.0~7.5之间,盐度为20~30左右;Bdh5221增殖速度与起始接种浓度影响不大,起始接种浓度10<'1>~10<'3>pfu/ml下经过5~7d生长可增殖4log左右;Bdh5221保藏选择20%~30%甘油含量较为合适,低温条件更利于保藏。 安全性试验结果表明,添加1.5×10<'6>pfu/ml蛭弧菌海水菌株Bdh5221对南美白对虾无毒害作用;添加2.0×10<'2>pfu/ml,2.0×10<'3>pfu/ml,2.0×10<'4> 16

pfu/mlBdh5221均对锯缘青蟹蚤状幼体阶段五个变态期幼体存活率和变态率无影响。锯缘青蟹苗种培育中应用蛭弧菌海水菌株Bdh5221试验,进一步表明,其最适有效添加量为1.5×10<'1>pfu/ml;添加蛭弧菌的试验组水体微生态环境明显改善,在育苗后期,相较对照组而言,试验组水体中弧菌和细菌总数都维持在较低水平,两者相差1~2个数量级,而成活率和变态率试验组则比对照组分别高6.6%和6.3%;同时试验组水质有所改善,其中NH<,4>-N比对照组低0.11~0.34 mg/1。 从蛭弧菌在锯缘青蟹苗种培育的应用试验中可以看出,其在海水苗种培育中维持微生态平衡,防治病害方面有广泛的应用前景。

Abstract

The addn. of Timsen (alkyl di-Me benzyl ammonium chloride-40-active) to a shrimp culture pond effectively prevented the occurrence of vibriosis in shrimp. The results indicated that Timsen could effectively reduce the no. of total heterotrophic bacteria in water samples, but its efficacy was reduced in brackish as compared with fresh water.

A significant redn. of the no. of Vibrio species in brackish water samples was also obsd. In the hepatopancreas of shrimp collected from a control pond, a rapid increase in the no. of Vibrio was obsd. with the onset of disease. With prolonged exposure (120 h), Timsen still maintained lower Vibrio nos. in brackish water samples, with no difference in total nos. of heterotrophic bacteria between control samples and treated brackish water samples. Further anal. of the compn. of the Vibrio species suggested that Timsen also caused a change in the components of the Vibrio community. Results also indicated that Timsen may affect the hemocytic activity of shrimp by enhancing the activity of phagocytosis and/or encapsulation; however, it also induces hemocyte death via apoptosis.

Abstract

AIMS: The present work aims at finding potential probionts from marine sources as a biocontrol agent against 17

pathogenic Vibrio species in shrimp larval culture. METHODS AND RESULTS: A total of 109 bacterial strains were isolated from seawater, sediment and marine fish-gut samples, and were screened for their antagonistic activity against Vibrio species. Three strains (Q, Q1 and M) isolated from the marine sediment were found antagonistic against Vibrio strains. Based on 16S ribosomal DNA gene sequence analysis, the strain Q was identified as Paenibacillus spp. (EF012164); Q1 as Bacillus cereus (DQ915582); and the M as Paenibacillus polymyxa (DQ915580). Further, the two bacterial species, Paenibacillus spp. and B. cereus were challenged separately at two different concentrations of 10(4) and 10(5) CFU ml(-1) for probiotic activity in the postlarvae of Penaeus monodon against pathogenic Vibrio harveyi and Vibrio spp. CONCLUSIONS: The present study identified the probiotic activity of Paenibacillus spp., B. cereus and Pa. polymyxa against the pathogenic Vibrios in the postlarvae of P. monodon. SIGNIFICANCE AND IMPACT OF THE STUDY: In vivo study reveals that the marine bacterial species can be used as probionts against pathogenic Vibrios in shrimp larval culture practices.

Abstract

Use of antibiotics for the control of bacterial diseases in shrimp culture has caused several adverse impacts to the industry. This has resulted in the search for alternative environment friendly approaches to overcome bacterial infections. This study was conducted to investigate the use of beneficial bacteria as an alternative to antibiotics. Ten pathogenic bacterial species isolated from shrimp, Penaeus monodon, and Artemia cysts were tested for susceptibility to indigenous marine Bacillus subtilis AB65, Bacillus pumilus AB58, Bacillus licheniformis AB69 and compared with oxytetracycline, chloramphenicol, gentamicin and bacitracin, which are common antibiotics used in Asian aquaculture. The Bacillus spp. were isolated from the local marine environment for bioremediation use in shrimp hatcheries and were proven to reduce total ammonium nitrogen. The pathogenic bacterial isolates were 90% susceptible to B. subtilis AB65, 70% susceptible to B. pumilus AB58 and B. licheniformis AB69 and 100% susceptible to oxytetracycline, chloramphenicol and gentamicin but only 40% to bacitracin. Two representative isolates of the vibrio group, Vibrio alginolyticus VaM11 and Vibrio parahaemolyticus VpM1, when tested for competitive exclusion by a common broth method using the marine Bacillus spp., showed decreased viable counts from 108 to 102 cfu mL-1. The results suggest that the action of the marine bacteria appears to be significant in protecting the host shrimp against pathogenic bacteria. In addn. to the alternative use of antibiotics, the selected 18

marine bacteria had addnl. bioremediation properties of reducing ammonia.

Abstract

Four marine Bdellovibrio-and-like organisms (BALOs), viz. BDW01, BDW02, BDW03 and BDW04, were isolated from the Shenzhen Bay of Guangdong province, China, using Vibrio parahaemolyticus (Vp) strain minus as the host bacterium. They were identified as BALOs by electron microscopy and specific polymerase chain reaction methods. Lysis expts. on 41 potentially pathogenic vibrios showed that individually, BDW01, BDW02, BDW03 and BDW04 lysed 36.6, 39.0, 65.8 and 63.4% vibrios, resp. In combination, they lysed 87.8% of the vibrios tested. In terms of different vibrio species lysed, four BALOs combined could lyse 81.8% Vibrio alginolyticus (11 strains), 83.3% non-O1 Vibrio cholerae (6 strains), 88.9% Vibrio fluvialis (9 strains), 100% Vibrio mimicus (4 strains) and 88.9% Vp (9 strains). Methods and Results: Using double-layer agar technique, the nos. of culturable BALOs that lyse Vibrio alginolyticus were found to be 10-103 PFU ml-1 in the surface water samples. A total of 130 BALOs isolates were differentiated into five phylotypes by denaturing gradient gel electrophoresis targeting the 16S rDNA V3 region and four phylotypes by amplified rDNA restriction anal. of the Bacteriovoracaceae-specific 16S rDNA fragment resp. Phylogenetic anal. of representative isolates showed that all of them were identified as Bacteriovorax spp., but affiliated with four different clusters in the family Bacteriovoracaceae. Conclusions: The two PCR-based methods both can be chosen to rapidly type the saltwater BALOs at cluster level. And the relatively large nos. of BALOs with various phylotypes recovered from the same habitats suggested that these predators might play important ecol. role in shrimp mariculture environments. Significance and Impact of the Study: We proposed two effective methods to distinguish rapidly large nos. of BALOs isolates and our results would be helpful to understand the diversity and function of BALOs in mariculture environments.

Also claimed is (I) treatment and prophylactic technique for Vibrio infectious disease, wherein the therapeutic agent is scattered in culture fishery water (pond) for a marine life; and (II) a treatment and prophylactic technique for Vibrio infectious disease, wherein the therapeutic agent is mixed with a feed and is administered to marine life in the culture fishery.

The microorganism is pref. Bdellovibrio stolpii esp. Bdellovibrio stolpii subsp. BD-4 (Bikokenki No. 12417).

USE/ADVANTAGE-The Vibrio infectious disease can be treated without generating resistant microbe nor damaging safety by using Bdellovibrio (i.e. Leptospira interrogans of Vibrio). Handling of the therapeutic agent is very easy and the cost is economical compared to conventional agent.

Intramuscular injection of Vibrio, Bdellovibrio and their mixt. (0.5 ml of a sterilised sea water suspension in Vibrio and Bdellovibrio (10 cells/ml)) was carried out on Peneus japonicus (11-13g) abdomen, and also only the sterilised sea water was injected as a control. Mixing rate of the Vibrio and Bdellovibrio was 9:1. The next day, all of the prawns (5) in Vibrio-injection area were dead, however, in the Vibrio-Bdellovibrio injection area, three were alive. In the control and Bdellovibrio injection areas, all 5 were alive.

Parasitic bacteria of the genus Bdellovibrio are utilized to control or eliminate undesirable bacteria in or on: certain foods for human consumption; or a food contact surface(s) (i.e. the Bdellovibrio attack and kill undesirable bacteria indigenous to the food or food contact surface(s)). A wide variety of pathogenic (e.g. Salmonella, Vibrio, Shigella, Escherichia, Campylobacter and Yersinia), and food spoilage bacteria (e.g. Pseudomonas, Aeromonas and Enterobacteriaceae) may be controlled or eliminated by parasitization by the Bdellovibrio bacteria.

1、课题信息总结

国内有蛭弧菌微生态制剂生产方法及应用的文献报道,提及应用于养殖水体、虾养殖环境、南美白对虾等等,但未见针对斑节对虾采用蛭弧菌微生态制剂控制的应用研究;有微生物制剂对斑节对虾亲虾池异养细菌的影响的文献报道,不是采用蛭弧菌微生态制剂控制。因此,国内公开发表的中文文献中,未见采用蛭弧菌微生态制剂作为控制手段,探讨斑节对虾海产品绿色安全养殖生产的文献报道。

国外有虾或者斑节对虾弧菌疾病控制和生物控制的文献报道(文献12~15),但未见涉及采用蛭弧菌微生态制剂控制。国外有蛭弧菌(或类生物)微生态制剂研究及应用于食品、水产、虾的文献报道(文献16~19),但未见针对斑节对虾控制弧菌数量的安全养殖的研究。

2、 检索经验总结:

(1)本课题检索,主要检索词为蛭弧菌、斑节对虾,但课题分析尽量提炼相关知识点。

(2)调整检索策略。一开始用很少的检索词限制在主题(题名、关键词、文摘)中,在选择的数据库快速搜一遍,根据检索结果多少,建立检索式,并不断尝试和调整。

(3)进行外文数据库检索,用检索式(Shrimp or prawn or "penaeus monodon") and bdellovibrio*先过一遍, 20

发现相关文献很少,基本上确认很多数据库只用一个词即可bdellovibrio*。但在CA或ISI数据库中,仅用bdellovibrio检索结果过多,需要增加一些主题概念加以限制,用逻辑“与(AND)”进行组配,还常常加入语种限制或分析,去掉中文部分。在挑选相关文献时,也很费工夫,检索式(Shrimp or prawn or "penaeus monodon") and bdellovibrio*相关文献很少,就要改变策略,分别围绕关键词为蛭弧菌用于食品(水产海产)、斑节对虾弧菌生物控制来挑选相关文献。

(4)使用搜索引擎检索,最好使用高级检索,可以做各种限制,检索词可以实施布尔逻辑组配。

(5)同一个检索式在不同数据库检索,必须注意每个数据库的检索界面、检索语言、检索策略实施均有不同。
